AI-designed drug Rentosertib shows biological age reduction in clinical trials
Insilico Medicine announced research published in Nature Biotechnology showing that its AI-designed drug candidate, Rentosertib (ISM001-055), significantly reduced age-related biomarkers in human clinical trials. Over a 12-week trial involving 43 patients with idiopathic pulmonary fibrosis, all six evaluated 'aging clocks' registered a measurable decline in predicted biological age. This is a pioneering milestone where an AI-generated small molecule targeting a novel biological pathway has demonstrated potential anti-aging effects in humans. It demonstrates how generative AI in biotechnology can expand beyond traditional short-term disease treatment into preventive longevity research. Rentosertib was created using AI models that scanned protein targets to find a novel pathway (TNIK inhibitor) for idiopathic pulmonary fibrosis and generated a matching molecular structure. However, experts emphasize that the anti-aging findings are preliminary, based on a small patient sample, and have not yet been tested in healthy individuals.
## BACKGROUND
Insilico Medicine uses generative artificial intelligence and neural networks to analyze biological data, identify novel disease targets, and design custom drug molecules. 'Aging clocks' are biological algorithms—often based on DNA methylation or blood biomarker patterns—that estimate cellular health and biological age relative to actual chronological age. Idiopathic pulmonary fibrosis (IPF) is a chronic, age-related lung disease causing progressive lung tissue scarring and diminished respiratory function.
